Potassium hydroxide and sodium hydroxide solutions were used as basic catalysts. Triacylglycerols were transesterified, rapeseed oil was used as a sample. Triacylglycerols were converted to methyl esters of fatty acids, which were subsequently determined by GC-MS where the peak areas were monitored, because they represent the yield of the formed fatty acid methyl esters, namely palmitic, oleic, linoleic and stearic acids. The highest yield of transesterification of the oil to methylesters of fatty acids was found for a concentration of 3.0 mol dm<sup>–3</sup> for KOH and 2.0&nbsp;mol&nbsp;dm<sup>–3</sup> for NaOH. At higher catalyst concentrations, a predominant side-reaction, saponification, was observed, which decreased the overall yield of fatty acid methyl esters.</p> Adam Březina Adéla Dořmanová Petr Kukučka Přemysl Lubal Marta Farková Copyright (c) 2022 2022-11-02 2022-11-02 20 5 319 322 10.54779/ccsss20220319 Využitie extrakcie podporenej ultrazvukom pre analýzu škorice a potravinových výrobkov http://www.ccsss.cz/index.php/ccsss/article/view/10 <p>The objective of this study was to select suitable experimental conditions for extraction of coumarin by Ultrasonic Assisted Extraction (UAE). Type of extraction solvent, extraction time and temperature were optimized to increase the extraction yield of coumarin from samples of cinnamon and foods containing cinnamon. Optimal UAE conditions include extraction time of 10 min, temperature of 50 °C and extraction solvent based on choline chloride and ʟ-lactic acid. The extraction yields were comparable to traditional solvents – water and methanol.</p> Kristína Čišovská Katarína Hroboňová Copyright (c) 2022 2022-11-02 2022-11-02 20 5 323 326 10.54779/ccsss20220323 Studium využitelnosti koloidních roztoků zlatých a stříbrných nanočástic pro povrchem zesílenou Ramanovu spektroskopii krevní plazmy http://www.ccsss.cz/index.php/ccsss/article/view/11 <p>This work focuses on the preparation of colloidal solutions of Au and Ag nanoparticles and verification of their applicability for surface-enhanced Raman scattering spectroscopy of blood plasma. The composition of blood plasma may change due to pathobiochemical processes associated with disease development. Analysis of blood plasma might reveal these changes and thus help in the diagnostics process of various diseases. Raman spectroscopy appears to be an ideal method for rapid and non-destructive analysis of blood plasma. Moreover, surface-enhanced Raman scattering spectroscopy might allow the detection of very low levels of potential disease-specific markers. In this work, the prepared colloidal solutions of Au and Ag nanoparticles were first characterized by UV‑VIS absorption spectroscopy and their enhancing capabilities were verified using riboflavin as a model analyte. Subsequently, optimization of the measurement conditions for surface-enhanced Raman spectroscopy of blood plasma was performed. The results show that at the excitation wavelength of 785 nm, significantly higher enhancement is achieved when using Ag nanoparticles. In the case of Au nanoparticles, the enhanced blood plasma spectrum was successfully detected only once, and the addition of an aggregating agent (NaCl) was required.</p> Jakub Harvalík Kateřina Hrubešová Ondřej Vrtělka Vladimír Setnička Copyright (c) 2022 2022-11-02 2022-11-02 20 5 327 330 10.54779/ccsss20220327 Vliv podkladového materiálu a přídavku komplexačního činidla na galvanickou přípravu SERS-aktivních vrstev a spektra riboflavinu http://www.ccsss.cz/index.php/ccsss/article/view/12 <p>SERS spectroscopy remains not a well-established analytical technique mainly because of the complicated preparation of SERS-active substrates, which often have low reproducibility. For this reason, the development of new or optimization of existing substrates is important. For testing the SERS activity of these substrates, the so-called model analytes are commonly used. One of them is riboflavin, which is utilized for its non-toxicity and suitable optical properties.</p> <p>This work is focused on optimizing the enhancing properties of galvanically prepared silver substrates. The influence of the base material (Al and its alloys, Zn) and of the addition of a complexing agent (EDTA) into the Ag deposition bath on the spectral response of riboflavin was evaluated. Each prepared substrate was characterized by scanning electron microscopy, and the SERS spectra were collected using an excitation wavelength of 1064 nm. All data were subsequently processed and interpreted using multivariate statistics, namely principal component analysis. The most universal and suitable base material for the preparation of Ag substrate for riboflavin detection was the AlMg alloy. The addition of a complexing agent to the deposition bath changed the morphology of the prepared substrates and the highest spectral response of riboflavin was obtained by adding 0.55 mM EDTA. The intensity of riboflavin spectral response was comparable to the value obtained using substrates prepared in the Ag bath, but the substrates prepared in the AgNO<sub>3</sub> solution achieved a&nbsp;weaker enhancement of the spectral response.</p> Adéla Koryťáková Oleksandr Volochanskyi Martin Král Marie Švecová Copyright (c) 2022 2022-11-02 2022-11-02 20 5 331 334 10.54779/ccsss20220331 Detekce proteinových biomarkerů pomocí lateral flow testů s foton-upkonverzními nanočásticemi http://www.ccsss.cz/index.php/ccsss/article/view/13 <p>Rapid and sensitive detection of clinically important analytes is crucial for early disease diagnosis and initiation of treatment. Immunochemical assays are widely used to determine the presence and concentration of analytes in various complex media due to their high specificity provided by antibodies. Recently, lateral flow immunoassays (LFIAs) are becoming one of the most popular forms of immunochemical assays. LFIAs are test strips based on nitrocellulose membranes, and their most significant advantage is that there is no need for laboratory equipment to perform the assay enabling their use for point-of-care testing. Most LFIAs are based on gold nanoparticles; however, they often limit the resulting sensitivity of LFIAs. Our work focused on photon-upconversion nanoparticles (UCNPs) as a sensitive alternative label. UCNPs are lanthanide-based nanocrystals exhibiting anti-Stokes luminescence (excitation by the NIR laser and detection in the Vis region), which allows detection without optical background interference. We have developed an LFIA assay for human serum albumin (HSA) with UCNPs as a label. Using a specific anti-HSA antibody and a control anti-mouse antibody, various LFIA parameters were optimized. The immobilization of antibodies was done using acetate buffer with 1% methanol, glass membrane ST17 was used as a conjugate pad, and Tris buffer with 5% sucrose was used to stabilize the particles within the conjugate pad. The choice of a different nitrocellulose membrane had a&nbsp;relatively small effect on assay performance; the best results were achieved with membrane 120 HP Plus Thick.
